Clostridium perfringens NetB toxin is a β-pore-forming toxin secreted by certain strains of the anaerobic bacterium Clostridium perfringens, specifically type G strains. NetB plays a critical role in the pathogenesis of avian necrotic enteritis, causing significant economic losses in the poultry industry. The toxin is produced as a soluble monomer that oligomerizes on the surface of susceptible host cells—primarily chicken hepatocytes—inserted into the membrane as a heptameric β-barrel pore with a diameter of ~1.6–1.8 nm. This pore formation leads to loss of cellular integrity, osmotic imbalance, and ultimately cell lysis. NetB is structurally related to staphylococcal alpha-hemolysin but differs in membrane specificity and mode of receptor binding, interacting preferentially with cholesterol-rich membranes rather than phosphocholine. The presence of the netB gene is a crucial biomarker of virulent C. perfringens strains in poultry disease. While no direct pharmacologic inhibitors are in clinical use, NetB is a target for vaccine development in animal health, and understanding its structural mechanism informs efforts to control necrotic enteritis in livestock.
