The clotting factor carboxylation pathway refers to the vitamin K-dependent post-translational modification of specific coagulation factors. This process is essential for the biological activity of these proteins, enabling them to bind calcium ions and participate effectively in the blood coagulation cascade. The central enzyme in this pathway is gamma-glutamyl carboxylase (GGCX), which catalyzes the addition of a carboxyl group to specific glutamic acid residues at the N-terminus of target proteins, converting them into gamma-carboxyglutamic acid residues. The main targets are several clotting factors: II (prothrombin), VII, IX, X, as well as regulatory proteins C, S, and Z. Gamma-carboxyglutamic acid residues enable these factors to bind calcium ions efficiently, allowing their association with phospholipid membranes.
