Club Cell Secretory Protein (CCSP), also known as Secretoglobin 1A1 (SCGB1A1), is a small, secreted homodimeric protein of approximately 16 kDa belonging to the secretoglobin superfamily. It is primarily produced by club cells in the terminal bronchioles of the lung. CCSP plays crucial roles in barrier protection, anti-inflammation, immunomodulation, antioxidation, detoxification, and epithelial repair/regeneration. It is clinically relevant as a biomarker for lung injury and a potential therapeutic target for respiratory diseases.
Direct binding via an LVD motif on α4β1 integrin/VLA‑4 on leukocytes—reducing their adherence/migration into inflamed lungs—and inhibition of TLR4/NF-kB signaling pathways that drive inflammation. It also limits PLA2 activity involved in generating pro-permeability oxidized lipids.
