Overview

CD154, also known as CD40 ligand (CD40L), is a type II transmembrane protein and a member of the tumor necrosis factor (TNF) superfamily. It plays a central role in immune regulation, particularly in T cell–B cell interactions and inflammatory responses. It acts as a costimulatory molecule essential for adaptive immunity, promoting B cell proliferation, inducing immunoglobulin class switching, supporting germinal center formation, facilitating memory B-cell generation and stimulating cytokine production. Elevated levels of soluble or membrane-bound CD154 have been associated with autoimmune diseases and cardiovascular disorders. Genetic defects affecting the interaction between this molecule and its main receptor result in severe immunodeficiency syndromes characterized by defective antibody responses.
