Overview

CD228/melanotransferrin is a transferrin family member highly expressed on several solid tumors but minimally present on most normal adult tissues. Its biological roles include promoting angiogenesis and possibly facilitating metastasis. These features make it an appealing target for novel immunotherapies—including ADCs and bispecific agents—currently under clinical investigation for multiple cancers.

Bispecific molecules like SGN-BB228 bind both CD228 on tumor cells and activate costimulatory receptors like 4‑1BB on T cells, leading to enhanced T-cell cytotoxicity localized at sites where tumor cells express high levels of CD228.
