Overview

Fatty acid translocase (CD36) is a membrane-associated transporter protein with high affinity for long-chain fatty acids. It plays a crucial role in facilitating the uptake and transport of these fatty acids into cells—particularly skeletal muscle and heart—and across mitochondrial membranes where they undergo β‑oxidation to produce energy. The presence and function of FAT/CD36 on both plasma and mitochondrial membranes are essential for efficient cellular fat utilization, especially during periods of increased metabolic demand such as exercise. Experimental inhibition with sulfo-N-succimidyl oleate dramatically reduces palmitate oxidation, highlighting its central role in regulating lipid-derived energy production. Alterations or deficiencies in FAT/CD36 expression have been linked to metabolic diseases including insulin resistance, obesity, cardiovascular disease, and may also influence tumor cell metabolism due to its impact on cellular fuel supply.
