CD47, also known as integrin-associated protein (IAP), is a ubiquitously expressed transmembrane glycoprotein that plays critical roles in cellular communication, immune regulation, and disease pathology. It interacts with SIRPα on macrophages to inhibit phagocytosis, acting as a "don't eat me" signal. It also interacts with thrombospondin 1 (TSP-1) and membrane integrins. Overexpression on tumor cells contributes to immune evasion, making it a target for anti-cancer therapies. CD47 also influences apoptosis, cell proliferation, and migration. Altered expression is implicated in diseases like Gaucher disease and pulmonary fibrosis.
