Overview

CD72 is a type II transmembrane glycoprotein expressed on B lymphocytes, acting as a co-receptor for the B cell receptor (BCR). It contains a C-type lectin-like domain and plays a role in regulating B cell signaling, primarily through its inhibitory function mediated by ITIM motifs and recruitment of SHP-1. CD72 recognizes RNA-containing self-antigens and is implicated in the pathogenesis of autoimmune diseases like SLE. It is also associated with chronic lymphocytic leukemia and may have prognostic value in kidney renal clear cell carcinoma.

Mechanism of action

CD72 acts as a co-receptor for the B cell receptor (BCR). Its cytoplasmic tail contains ITIM motifs that, upon phosphorylation, recruit SHP-1, leading to downregulation of BCR signaling and inhibition of B cell activation. This mechanism helps maintain self-tolerance, particularly against RNA-containing self-antigens.
