Overview

Clusterin (CLU), also known as Apolipoprotein J, is a multifunctional, secreted glycoprotein that acts primarily as an extracellular molecular chaperone. It is widely expressed and found in various body fluids. Clusterin binds misfolded proteins, inhibits apoptosis, and prevents complement-mediated cell lysis. Alterations in clusterin expression are implicated in numerous diseases, including Alzheimer's disease and cancer.

Mechanism of action

Acts as an ATP-independent molecular chaperone for misfolded proteins; inhibits apoptosis by interacting with BAX on mitochondria and modulating survival pathways; prevents complement-mediated cell lysis by binding terminal complement complexes.
