Overview

Clusterin pre-messenger RNA is the unspliced transcript of the clusterin gene, which undergoes alternative splicing to generate different mRNA isoforms. These transcripts, after splicing, produce protein variants with diverse functions, including roles in apoptosis and tumorigenesis. Aberrant splicing of clusterin pre-mRNA has been implicated in cancer development, cell survival, and therapy resistance. While the mature Clusterin protein and its isoforms are therapeutic targets, the pre-mRNA itself is not conventionally targeted, but modulation of its splicing by spliceosome-targeting drugs or antisense approaches may impact the function of Clusterin protein in disease contexts[6]. Clusterin pre-mRNA is not itself a canonical therapeutic target but is critical for the regulation of Clusterin protein isoforms via alternative splicing. Targeting mechanisms that affect its splicing may offer therapeutic potential, primarily by altering the balance of functional Clusterin isoforms, which are implicated in cancer cell proliferation, apoptosis, and metastasis[6].
