Overview

Cms1 ribosomal small subunit homolog (CMSS1) is a conserved human RNA-binding protein implicated in ribosome assembly and RNA metabolism. CMSS1 is associated with the biogenesis of ribonucleoprotein complexes, rRNA processing, ncRNA metabolism, translation initiation, and RNA localization. Structurally, it exhibits a helicase fold, although it likely lacks classic helicase activity. CMSS1 is dynamically involved in 90S pre-ribosome maturation and modulates the timing of snoRNA-guided rRNA modification, particularly in the maturation of the small ribosomal subunit. Elevated expression of CMSS1 has been linked to poorer clinical outcomes in cancers such as liver hepatocellular carcinoma and diffuse large B-cell lymphoma, implicating it as a potential molecular marker for cancer prognosis and immune microenvironment modulation. CMSS1 is also involved in maintaining HIV-1 latency, with loss of function leading to reversal of viral latency, suggesting a broader regulatory role in gene expression and cellular states[1][2][3][5].
