Overview

The physiological plasma coagulation and anticoagulation systems represent a complex, highly regulated network of proteins and enzymes that maintain blood fluidity while preventing excessive blood loss upon vascular injury. The coagulation cascade involves a series of zymogen activations, primarily serine proteases, culminating in the conversion of fibrinogen to fibrin to form a stable clot (StatPearls, 2023). This process is balanced by the anticoagulation system, which includes inhibitors like antithrombin, protein C, and protein S, ensuring that clot formation remains localized and does not obstruct normal blood flow (NIH, 2022). Dysregulation of this balance leads to either pathological thrombosis, such as deep vein thrombosis or stroke, or bleeding disorders like hemophilia. Pharmacological intervention typically targets specific components of this system, such as Factor Xa or thrombin, to treat or prevent cardiovascular and thromboembolic diseases (PubMed, 2021). While effective, these drugs require careful monitoring due to the inherent risk of major hemorrhage associated with suppressing the body's natural clotting ability.
