The coagulation cascade complex refers to the highly regulated and interlinked network of enzymatic reactions in blood plasma that produce a fibrin clot at the site of vascular injury. The process involves sequential activation (often by limited proteolysis) of zymogen coagulation factors via two initiating pathways—the intrinsic and extrinsic—which merge into a common pathway, ultimately generating thrombin and converting fibrinogen to cross-linked fibrin[1][2][3][5][8]. The cascade is central to normal hemostasis but can be pathologically dysregulated in both bleeding and clotting disorders[1][4][5][8]. It is not a single molecule but a dynamic series of multi-subunit protein complexes, including factors such as Factor VIIa/tissue factor (extrinsic complex), Factor IXa/VIIIa (intrinsic complex), and Factor Xa/Va (prothrombinase complex), as well as the final fibrin-stabilizing complex (Factor XIIIa)[1][5]. Therapeutic modulation of this cascade is the basis for many anticoagulant and procoagulant drugs, but interventions are associated with risks including bleeding and thrombosis[1][5][8].
