Overview

Coagulation factors are a group of essential plasma proteins that interact in a complex biochemical cascade to facilitate blood clotting and maintain hemostasis. Most of these factors are serine proteases that circulate as inactive zymogens and are sequentially activated through limited proteolysis in response to vascular injury (StatPearls, NBK507795). The cascade is organized into the intrinsic, extrinsic, and common pathways, ultimately leading to the conversion of prothrombin to thrombin, which then transforms soluble fibrinogen into an insoluble fibrin mesh (NIH, Hemophilia). Deficiencies in these factors, such as Factor VIII or IX, result in bleeding disorders like Hemophilia A and B, while overactivity or stasis can lead to life-threatening thrombosis (UniProt, P00734). These proteins are major therapeutic targets; anticoagulants like rivaroxaban and dabigatran inhibit specific factors to prevent stroke and venous thromboembolism, whereas purified or recombinant factors are administered as replacement therapy for bleeding disorders (PubChem, Rivaroxaban). Monitoring the activity of these factors is critical in clinical practice, often utilizing standardized tests like PT/INR and aPTT to ensure therapeutic efficacy and safety (Wikipedia, Coagulation factor).
