Overview

Coagulation factor IXa (UniProt P00740) and Coagulation factor XIIa (UniProt P00748) are two distinct serine proteases within the intrinsic and contact activation pathways of the blood coagulation cascade. Factor XIIa, also known as Hageman factor, initiates the contact pathway upon interaction with negatively charged surfaces and is a key link between coagulation and inflammation via the kinin-kallikrein system (StatPearls: Physiology, Coagulation Cascade). Factor IXa, or Christmas factor, is a vital component of the intrinsic tenase complex, which is responsible for the large-scale activation of Factor X required for fibrin clot formation (UniProt P00740). While Factor IXa is essential for normal hemostasis, Factor XIIa is considered a promising target for safe anticoagulation because its inhibition appears to prevent pathological thrombosis without significantly increasing bleeding risk (PMID 32810011). Drugs targeting these factors include monoclonal antibodies like Garadacimab for FXIIa (NCT04656535) and aptamers like Pegnivacogin for FIXa (PMID 25208214). These therapies are primarily developed for the prevention of stroke, venous thromboembolism, and the management of hereditary angioedema. Overall, these enzymes represent critical nodes for modulating blood clot formation and inflammatory responses in various cardiovascular and hematological diseases.
