The intrinsic tenase complex is a critical component of the intrinsic pathway of the blood coagulation cascade. It consists of activated coagulation factor IX (factor IXa), its cofactor factor VIIIa, calcium ions (Ca²⁺), and negatively charged phospholipid surfaces—typically provided by activated platelets. This complex catalyzes the activation of factor X to Xa, a key step leading to thrombin generation and fibrin clot formation. Factor VIIIa acts as an essential non-enzymatic cofactor, dramatically increasing FIXa's catalytic efficiency toward FX. Deficiencies or dysfunctions in any component—especially FVIII or FIX—result in hemophilia A or B respectively, characterized by impaired clotting.
