Coagulation proteases are a specialized group of enzymes, predominantly serine proteases, that play a central role in the blood coagulation cascade to maintain hemostasis (StatPearls, 2023). These proteins typically circulate as inactive zymogens and are sequentially activated through limited proteolysis in response to vascular injury (UniProt, 2024). The cascade involves two primary pathways—the intrinsic and extrinsic pathways—which converge on the common pathway to activate Factor X and subsequently Thrombin (Factor IIa) (NIH, 2023). Thrombin then converts soluble fibrinogen into insoluble fibrin, forming the structural mesh of a blood clot (PubMed, 2022). Pathological activation of these proteases leads to thrombotic disorders such as deep vein thrombosis, pulmonary embolism, and stroke, while deficiencies result in bleeding disorders like hemophilia (Mayo Clinic, 2024). Therapeutic intervention often involves anticoagulants that either directly inhibit specific proteases like Factor Xa and Thrombin or indirectly reduce their activity by antagonizing Vitamin K, which is essential for the post-translational modification of several key factors (DrugBank, 2024).
