Coatomer subunit delta (ARCN1) is a highly conserved protein and a core component of the COPI (Coat Protein Complex I) coatomer complex, which functions in vesicular trafficking—primarily in retrograde transport from the Golgi apparatus to the endoplasmic reticulum as well as intra-Golgi trafficking. ARCN1 is essential for maintaining efficient protein transport, glycosylation, and compartmentalization of secretory organelles. Mutations or disruption of ARCN1 impairs type I collagen transport, causes endoplasmic reticulum stress, and is linked to craniofacial and skeletal syndromes, neurodegenerative phenotypes, and modulation of cancer risk. Cellular localization studies reveal ARCN1 at both the Golgi and endoplasmic reticulum, often in complex with β-COP; it may also serve independent functions outside the classical COPI complex. ARCN1 expression is regulated by microRNAs and is involved in additional physiological and pathophysiological processes, including responses to viral infection and metabolic regulation.
