Coatomer subunit epsilon (COPE) is one of several subunits (alpha, beta, beta', gamma, delta, epsilon, and zeta) that make up the cytosolic coatomer protein complex I (COPI). COPI mediates retrograde vesicle trafficking between the Golgi apparatus and endoplasmic reticulum as well as intra-Golgi transport. COPE binds to dilysine motifs and is essential for vesicle formation from Golgi membranes, ER-Golgi protein transport, and maintaining organelle integrity. While crucial for cell function, mutations or disruptions in COPE impair vesicle formation, receptor recycling, and cause organelle dysfunction. However, COPE is not currently considered a drug target and does not have direct disease associations beyond rare genetic or transport-related disorders
