Overview

Cobalamin (vitamin B12) is the chemically complex, water-soluble vitamin essential for DNA synthesis, fatty acid and amino acid metabolism, and neurological function[1][3][5]. It acts as a cofactor for methionine synthase and methylmalonyl-CoA mutase, enabling crucial methylation and rearrangement reactions necessary for normal cellular function. Cobalamin analogs include naturally occurring and synthetic cobamides (such as cobinamide, hydroxocobalamin, methylcobalamin, and cyanocobalamin). Several analogs are used therapeutically to correct deficiencies or as antidotes for cyanide poisoning by directly binding toxins[2][4]. The molecule is absorbed in the gut using intrinsic factor and transported via transcobalamin proteins to target tissues[1][3][5]. Some rare inherited disorders affect cobalamin metabolism and are the subject of research with cobalamin analogs, but the family as a whole is not a conventional drug target in the manner of receptors, enzymes, or transporters. The use of "Cobalamin/Cobalamin analog behavior" as a target is therefore imprecise and unsuitable for structured target-oriented medicinal databases.

Mechanism of action

When administered as an antidote, hydroxocobalamin and some cobalamin analogs bind strongly to toxins (e.g., cyanide, H₂S), neutralizing them and allowing excretion as nontoxic complexes. As a vitamin/cofactor: serves as an essential coenzyme for specific metabolic reactions, especially in DNA synthesis and certain metabolic pathways.
