Overview

Coccidioides fungus antigens are a collection of immunogenic molecules derived from the dimorphic fungi Coccidioides immitis and Coccidioides posadasii, which cause coccidioidomycosis or Valley Fever (NIH: National Institute of Allergy and Infectious Diseases). These antigens, including the well-characterized Antigen 2/Proline-Rich Antigen (Ag2/PRA), are primarily located in the fungal cell wall and are essential for the transition between the environmental mold form and the parasitic spherule form (PubMed: 10858233). They serve as the primary targets for the host's adaptive immune system, particularly triggering Th1 and Th17 cellular responses which are crucial for containing the infection (PubMed: 25139887). In clinical practice, these antigens are utilized in diagnostic assays to detect specific antibodies and in skin tests, such as those using Coccidioidin or Spherulin, to evaluate cellular immunity (StatPearls: Coccidioidomycosis). While not traditional pharmacological targets for small-molecule inhibitors, they are the central focus of vaccine development aimed at providing long-term prophylaxis for individuals in endemic regions (PubMed: 33434153).
