COF-01 is a novel, proprietary therapeutic target identified through proteomic analysis and immunohistochemistry of patient-derived tumor samples by Champions Oncology. It is significantly overexpressed at both the protein and mRNA levels in various malignancies, including non-small cell lung cancer, head and neck cancer, and pancreatic cancer, where its expression correlates with poor overall survival. As a tumor-associated antigen, COF-01 is being targeted using next-generation base-modified ssDNA aptamers, known as Raptamers, developed by Raptamer Therapeutics and its spin-out Radiomer Therapeutics. These Raptamers are conjugated to potent cytotoxic agents like monomethyl auristatin E (MMAE) to form Raptamer-drug conjugates (Rap-DCs) or to radionuclides for targeted radiopharmaceutical therapy. These modalities selectively induce cell death in COF-01-positive cancer cells while sparing normal tissue, offering a potential precision medicine approach for high unmet medical need populations.
