Cohesin subunit SA-1 (encoded by the STAG1 gene, also known as Stromal antigen 1) is a key component of the cohesin complex, which is essential for sister chromatid cohesion, homologous recombination, and chromosome architecture via DNA looping. The cohesin complex is composed of SMC1, SMC3, RAD21, and either SA-1 or SA-2 in somatic cells. SA-1 stabilizes the complex via RAD21 interaction; its roles include regulation of cohesion loading and release, and telomere cohesion. Disruption of STAG1 function leads to severe developmental defects and is implicated in diseases including certain cancers and chromosomal disorders due to its pivotal role in genome integrity.
