Overview

Cohesin subunit SA-3 (STAG3) is a meiosis-specific component of the cohesin complex, a multi-subunit protein machinery essential for the stable cohesion of sister chromatids after DNA replication, especially during meiosis[6]. This subunit is uniquely expressed in gametogenic cells and is crucial for homologous chromosome pairing, synapsis, recombination, and segregation in both spermatogenesis and oogenesis[4][5][7]. Loss-of-function mutations in STAG3 cause infertility in both sexes due to meiotic arrest, disrupted chromosome axis formation, and synapsis defects. Beyond its canonical role in meiosis, recent studies suggest that STAG3 may also regulate exit from pluripotency in stem cells via post-transcriptional mechanisms, expanding its functional repertoire beyond chromosome cohesion[2]. STAG3 is not currently a therapeutic target but is an important diagnostic marker for certain heritable infertility syndromes[3].
