Overview

Coiled-coil and C2 domain-containing protein 1A (CC2D1A) is a highly conserved protein encoded by the CC2D1A gene. It functions as a transcriptional repressor, originally identified for repressing the serotonin receptor 1A gene in neurons. CC2D1A also acts as a scaffold protein, anchoring components of the PI3K/Akt pathway and influencing signal transduction and cell survival pathways. In cancer biology, elevated CC2D1A expression correlates with poor prognosis and resistance to chemotherapy in ovarian cancer, mechanistically contributing to survival signaling and reduced apoptosis. Additionally, inherited mutations in CC2D1A cause autosomal recessive intellectual disability, consistent with its neuronal roles[1][2][3].

02

Mechanism of action

Chemoresistance promotion through modulation of apoptosis and survival pathways, potentially via NFκB and Akt pathway scaffolding[1][2] Transcriptional repression of the serotonin receptor 1A gene in neuronal cells[3]

03

Biological functions

Transcriptional repression (notably represses serotonin receptor 1A gene expression in neuronal cells)Signal transduction (acts as a scaffold in the EGFR-PDK1/Akt pathway)Chemoresistance modulation
04

Disease associations

Cancer (particularly ovarian cancer chemoresistance)Neurodevelopmental disorders (autosomal recessive non-syndromic intellectual disability)
05

Safety considerations

Potential for drug resistance if CC2D1A is highly expressed, especially in ovarian cancer (reduced efficacy of chemotherapeutics)[1][2]Not directly targeted by approved drugs; off-target effects or cognitive side effects possible if targeted due to its role in the brain
06

Interacting drugs

Biomarkers

CC2D1A protein expression as a prognostic marker for poor survival and chemotherapy resistance in ovarian cancer[1][2]
