Overview

Coiled-coil domain containing 198 (CCDC198) is a protein encoded by the CCDC198 gene in humans, also known as factor associated with metabolism and energy (FAME) and C14orf105. It is a 296-amino-acid protein with predicted coiled-coil domains, expressed primarily in the kidney, liver, and pancreas, with additional expression in other tissues including the fallopian tubes and bladder. The protein localizes to plasma membranes and cytoplasmic vesicles, is involved in cellular transport and possibly membrane/exosome trafficking, and functions in energy homeostasis and iron metabolism—supported by interactions with proteins such as ferritin heavy chain 1 and molecules related to cell cycle and mitochondrial function. Experimental knockout models show effects on body weight, lean mass, and serum ferritin, but without major overt pathology, suggesting molecular impacts on metabolism and excretion. CCDC198 has been proposed as a biomarker for liver fluke infection and alcoholic hepatitis, and very limited genetic evidence links certain polymorphisms to kidney disease and metabolic traits, though it is not an established therapeutic target and has no known drug interactions.
