Overview

Coiled-coil domain containing 62 (CCDC62) is a protein primarily functioning as a nuclear receptor coactivator that enhances the transcriptional activity of estrogen receptors (ESR1 and ESR2), and to a lesser extent modulates other nuclear hormone receptors including progesterone, glucocorticoid, and androgen receptors[1][3]. It plays a critical role in the positive regulation of transcription and in spermatid development, with high expression in the testis and a localization to sperm acrosomal vesicles and nuclei[1][3]. Loss-of-function mutations in CCDC62 in mice lead to defective acrosome formation in sperm and male infertility[1]. Genetic studies have implicated variants in CCDC62 as risk factors for Parkinson's disease, implicating it in neurodegeneration, and changes in its expression may be relevant to hormone response in cancer (e.g., prostate cancer)[1][3]. There are currently no approved drugs known to specifically target CCDC62, although it has been shown that fluoxetine can upregulate its transcription[1].
