Coiled-coil domain-containing protein 102B (CCDC102B) is a structural protein that participates in the assembly and function of the centrosome linker, which is essential for maintaining centrosome cohesion in interphase cells[1][3]. CCDC102B is recruited to the centrosome by the protein C-Nap1 (CEP250) and interacts with key linker components such as rootletin and LRRC45, facilitating the organization of rootletin filaments and thereby supporting centrosome stability[1][3]. Phosphorylation by the kinase Nek2A regulates its association with the centrosome, particularly during cell cycle transitions[1]. CCDC102B is also implicated in human disease. It is upregulated in metastatic breast cancer, where it promotes cell migration, invasion, and metastasis by modulating the NF-κB pathway; high levels are correlated with worse clinical outcomes[2]. In addition, genetic variants of CCDC102B have been associated with increased risk for myopic maculopathy, especially in individuals with high myopia[3]. The methylation status of CCDC102B serves as a molecular biomarker for biological age estimation in forensic applications[3]. Despite these emerging roles, CCDC102B is not considered a classical therapeutic target such as a receptor, enzyme, or transporter; there are currently no known drugs that directly act on or are developed to modulate this protein. Its significance is primarily structural and regulatory in centrosome biology, with recent interest in its relevance to cancer progression, eye diseases, and aging[1][2][3].
