Overview

Coiled-coil domain-containing protein 106 (CCDC106) is a 280-amino acid, ~32 kDa protein that contains a prominent coiled-coil domain, harbors a bipartite nuclear localization signal, and can directly interact with the transactivation domain of p53 in the nucleus[1][3][4]. CCDC106 promotes p53 protein degradation, thereby inhibiting p53-dependent transcriptional activity and supporting cancer cell proliferation and survival[1][3][4][7]. Additionally, CCDC106 can suppress p21 transcription through a p53-independent pathway involving interaction with ATF4, further enhancing cell proliferation, invasion, and EMT—particularly in mutant p53 ovarian cancer cells[2][5][6][8]. Overexpression of CCDC106 is linked to tumor progression and poor prognosis in multiple cancers[1][2][5][6][8]. Its subcellular localization (nuclear vs. cytoplasmic) modulates its biological activity, and nuclear CCDC106 is required for cancer-driving effects[2]. There is no indication that CCDC106 is misspelled or otherwise an incorrect target. No major interacting therapeutic drugs are currently known, but CK2 inhibitors can disrupt CCDC106 nuclear activity and may be explored as antitumor agents[1][2].
