Overview

Coiled-coil domain-containing protein 116 (CCDC116) is a protein encoded by the CCDC116 gene located on human chromosome 22q11.21. The protein is 612–613 amino acids long and contains a coiled-coil domain, but its precise molecular and cellular functions remain poorly characterized[2][5][7]. CCDC116 localizes to the centrosome, implicating a role in centrosomal architecture or function[1]. Immunohistochemical data show expression in most insulin-producing pancreatic islet cells, somatostatin cells, and pancreatic polypeptide cells, but not in glucagon or ghrelin cells, suggesting possible involvement in endocrine regulation and the biology of endocrine pancreatic tumors[7][1]. The gene has been investigated for potential associations with various cancers such as lung and prostate cancer, with genetic studies identifying variants linked to lung cancer susceptibility[1][2][5]. Proteomic and expression profiling also suggest a role in testis and potentially in ciliary biology[5]. However, there are no known drugs targeting CCDC116, and neither a therapeutic nor a validated biomarker role has been established to date[1][5][7].
