Overview

Coiled-coil domain-containing protein 12 (CCDC12) is a structural protein containing a coiled-coil domain, encoded by the CCDC12 gene on chromosome 3p21.31. While its precise molecular function remains incompletely characterized, CCDC12 plays a significant role in the development and progression of colon adenocarcinoma, where it is consistently overexpressed in tumor tissue compared to normal controls. Knockdown of CCDC12 in colon cancer cells reduces proliferation, migration, invasion, and tumorigenicity, and increases apoptosis, indicating its oncogenic potential. It is evolutionarily conserved and has been predicted (in zebrafish and other models) to participate in mRNA splicing within the U2-type spliceosomal complex. Like other coiled-coil proteins, CCDC12 is likely involved in facilitating protein-protein interactions and intracellular scaffolding. Current literature does not establish CCDC12 as a classic therapeutic target such as a receptor, enzyme, transporter, or ion channel, nor are drugs or inhibitors described that specifically target it at this time.
