Overview

Coiled-coil domain-containing protein 120 is a protein encoded by the CCDC120 gene, which includes two short coiled-coil domains at the N-terminus and a long proline-rich domain at the C-terminus[2]. CCDC120 localizes to the subdistal appendages of mother centrioles and proximal ends of both centrioles during the cell cycle, and is crucial for anchoring microtubules in interphase cells[2]. It cooperates with other subdistal appendage components such as ODF2, Ninein, and CEP170 to mediate the hierarchical assembly of these structures and recruits Ninein and CEP170 to the centrosome[2]. In addition, CCDC120 is required for neurite growth by interacting with cytohesin-2 to facilitate vesicular transport and ARF6 activation in neuronal cells[2][1]. Several alternatively spliced transcript variants encoding multiple isoforms of CCDC120 have been identified[1]. While there are disease associations based on genetic studies (e.g., differentiating neuroblastoma, Seckel syndrome 7), no clear functional disease mechanisms, validated drug interactions, or biomarker applications are reported. CCDC120 is, therefore, best classified as a centrosome scaffolding and structural protein, not a therapeutic target[1][2].
