Overview

Coiled-coil domain-containing protein 134 (CCDC134) is a secreted and intracellular protein characterized by a coiled-coil domain. It functions as a cytokine-like molecule that enhances CD8+ T cell-mediated immunity and exhibits antitumor and anti-inflammatory effects. CCDC134 also operates as an ER-resident chaperone, ensuring proper maturation and trafficking of TLR4 by regulating gp96 glycosylation, and may participate in chromatin modification through interaction with the p300-CBP-associated factor complex. Deficiency of CCDC134 is embryonic lethal in mice, underscoring its essential role in development and tissue homeostasis. Its expression is implicated as a prognostic biomarker in breast cancer and genetic variants are associated with osteogenesis imperfecta. No direct pharmacological agents are currently reported to target CCDC134, but it remains a potential immune and cancer therapeutic target due to its role in T cell and receptor signal modulation.
