Overview

Coiled-coil domain-containing protein 146 (CCDC146) is a microtubule-associated protein present in both somatic and germ cells, with crucial roles in centrosomal and axonemal structures[1][3][4]. In somatic cells, it localizes to the centrosome, mitotic spindle, and microtubule organizing center, suggesting involvement in cell division and spindle checkpoint control[1][3]. In spermatozoa, CCDC146 is a component of the flagellum and essential for spermatogenesis, with its loss causing male infertility in mice and humans due to immotile sperm[3][4]. CCDC146 contains multiple coiled-coil domains and a PF05557 mitotic checkpoint motif[1][3]. Its expression is associated with worse prognosis in several cancers including thyroid and breast, and it is considered a hub gene in these pathologies[2]. Although classified as a potential therapeutic target due to its disease associations, CCDC146 is currently considered difficult to drug via traditional small molecules, with no active inhibitors identified[4]. Alternative approaches like antisense oligonucleotides have been explored in translational research[4].
