Overview

Coiled-coil domain-containing protein 15 (CCDC15) is a centriolar protein associated with the inner scaffold of centrioles, a core structure essential for the size, length, and structural integrity of centrioles[2][4]. CCDC15 colocalizes and interacts with other inner scaffold proteins, such as POC1B and components of the SFI1/Centrin-2 complex, and is required for correct recruitment of these factors during centriole biogenesis and maintenance[2][4]. Loss of CCDC15 leads to defects in centriole length and structure, impairing the assembly and stability of primary cilia and reducing the ability of cells to respond to compartmentalized signaling such as Hedgehog signaling[2][4]. While coiled-coil domain proteins, in general, participate in a wide range of protein-protein interactions and structural cell processes, CCDC15 is specifically implicated in ensuring centriole size control, architectural integrity, and by extension, cellular development and health[2][4][3]. There are currently no known drugs that target CCDC15, nor is it considered a classical therapeutic target such as a receptor, enzyme, or transporter[2][4].
