Overview

Coiled-coil domain-containing protein 159 (CCDC159) is a structural protein enriched in spermatids, where it specifically localizes to the sperm head-tail coupling apparatus (HTCA)[1][2]. It contains a C-terminal coiled-coil domain that directs its localization to centrosomal structures. Experimental knockout of CCDC159 in mice results in acephalic spermatozoa (sperm lacking heads), abnormal flagellar development, and male infertility, underscoring its essential role in spermatid maturation and proper attachment of the sperm flagellum to the nuclear envelope[2]. CCDC159 may regulate this process by modulating the activity of protein phosphatase PP1 and is required for the correct structural assembly of the HTCA. There are currently no known therapeutic drugs or targeting agents for CCDC159, and it has not been described as a druggable therapeutic target in current biomedical literature[1][2][5][6]. References to coiled-coil domain-containing proteins in other contexts (such as CCDC3) do not apply to CCDC159[4]. - CCDC159 is a protein-coding gene (located on human chromosome 19), and not a member of common drug-target families such as receptors, enzymes, ion channels, or transporters[1][3]. - No known role in cancer, inflammation, neurodegeneration, or other major disease categories except male infertility[1][2]. - No clinical or experimental drugs, biomarkers, or safety/toxicity data are available for CCDC159. - No evidence this is a misspelled or incorrect target name[1][2].
