Overview

Coiled-coil domain-containing protein 186 (CCDC186) is a protein encoded by the CCDC186 gene on human chromosome 10q25.3[2][4]. This protein is primarily involved in the biogenesis and maturation of dense-core vesicles (DCVs), which are crucial for neuromodulator and hormone release in neurons and endocrine cells[1][3][7]. CCDC186 acts as a Rab2-interacting protein, participates in the trafficking and cargo sorting of secretory vesicles, and colocalizes with endosomal and recycling machinery[1][3][4]. Disruption or mutation of CCDC186 can impair vesicular trafficking, resulting in defective secretion of regulatory molecules, which may be relevant in both neurodevelopmental and endocrine pathology[1][7]. The gene is ubiquitously expressed with higher expression in some cancers (notably breast, bladder, and prostate), and has been implicated as a fusion partner in rare cancers[2][3]. CCDC186 is not a classical therapeutic target (not a receptor, enzyme, or transporter), nor are approved drugs known to target it directly[6][3].
