Coiled-coil domain-containing protein 25 (CCDC25)

Coiled-coil domain-containing protein 25 (CCDC25) is a highly conserved, 208-amino-acid human protein encoded on chromosome 8p21.1. It is expressed in a wide range of tissues and localizes mainly to the cell membrane and cytoplasm. Functionally, CCDC25 acts as a transmembrane receptor for DNA associated with neutrophil extracellular traps (NETs). Upon binding to NET DNA, CCDC25 activates the ILK-PARVB signaling pathway, leading to actin cytoskeleton rearrangement and enhanced cell migration. This mechanism is especially implicated in promoting tumor metastasis: NETs in the tumor microenvironment use CCDC25 to attract circulating tumor cells and encourage metastatic spread[2][3][6]. Beyond metastasis, CCDC25 is upregulated and released by tumor cells in cholangiocarcinoma and can be detected in serum, where its levels effectively discriminate malignant from benign and healthy states, showing potential as a high-sensitivity, high-specificity diagnostic biomarker[4][6]. The broad biological roles of CCDC25 are still being elucidated; however, its established role in cancer biology and emerging utility as a biomarker designate it as a promising therapeutic and diagnostic target.
