Coiled-coil domain containing protein 28A (CCDC28A) is a ubiquitously expressed, coiled-coil domain-containing protein of currently unknown definitive function in humans, encoded by the CCDC28A gene on chromosome 6[1][3][8]. The protein exists as two main isoforms distinguished by their N-termini, both containing a conserved coiled-coil motif, suggesting roles in protein–protein interactions but no known catalytic, transporter, or receptor activity[2]. While CCDC28A has no characterized function or biological pathway, research indicates potential involvement in discrete stages of hematopoietic development and in sperm morphology, particularly in head–tail coupling structure formation[2][4]. The gene is of interest in oncology because recurrent chromosomal translocations involving NUP98 and CCDC28A (forming the NUP98-CCDC28A fusion) have been identified in human acute leukemias, where the fusion protein induces proliferative and self-renewal capacities in myeloid progenitors in experimental models, although CCDC28A itself (without fusion) is not a known oncogene or therapeutic drug target[2][9]. There are no reported drugs that directly target CCDC28A, and there are no known mechanisms of action or biomarkers involving this protein in therapeutic contexts. Its biological and pathological functions remain largely uncharacterized.
