Coiled-coil domain-containing protein 28B (CCDC28B) is a protein localized mainly to centrosomes and basal bodies in cells, where it is involved in ciliogenesis—specifically, the formation and regulation of cilia length[1][2][3][4][5]. CCDC28B interacts with proteins associated with Bardet-Biedl syndrome (BBS) and acts as a genetic modifier in this context, with reduced CCDC28B levels correlating with more severe forms of the syndrome[1][3][4][5]. It participates in cilia biology by interacting with protein complexes such as mTORC2 via MAPKAP1/SIN1 and the kinesin 1 molecular motor, thereby impacting cilia length and subcellular distribution. CCDC28B on its own does not appear to be a direct cause of BBS or other classic ciliopathies in mouse models, but modifies their phenotypic expression when combined with other genetic mutations[4]. No evidence supports its use as a direct therapeutic target, biomarker, or drug-interacting molecule to date[5].
