Overview

Coiled-coil domain-containing protein 3 (CCDC3) is a secreted protein characterized by coiled-coil motifs, acting primarily as a regulatory factor in various cellular signaling networks. It is involved in protein-protein interactions, organization of cell structure, and important signal transduction pathways[1][2]. CCDC3 inhibits TNF-alpha-induced pro-inflammatory responses in endothelial cells by blocking NF-κB activation[2][6], and it modulates lipid metabolism by repressing inflammatory signaling and positively regulating lipid accumulation in adipose cells[2][7]. Aberrant regulation of CCDC3 has been implicated in cancer biology (by suppressing breast cancer growth through p53 stabilization[5]), metabolic disorders, and cardiovascular disease, suggesting a role as a disease biomarker and a potential (though not established) therapeutic target[1][2][5]. While not a classical receptor, enzyme, or transporter, CCDC3 is of marked interest as a secretory regulatory molecule in multiple disease contexts.
