Overview

Coiled-coil domain-containing protein 33 (CCDC33) is a protein encoded by the CCDC33 gene and contains several coiled-coil domains, a C2 domain, endoplasmic reticulum retention motifs, and peroxisomal targeting signals, directing the protein primarily to the peroxisome in male germ cells[1][2]. CCDC33 is predominantly expressed in testis, specifically in primary spermatocytes, suggesting a critical role in spermatogenesis[2][5]. It has also been identified as a cancer/testis antigen (CT antigen), indicating its potential relevance in certain cancers[2][5]. Genetic variations in CCDC33 have been correlated with increased susceptibility to invasive pneumococcal meningitis and altered expression observed in neurodegenerative contexts, such as Huntington’s disease[1]. The protein is multifunctional, implicated in both normal cellular processes and various disease contexts, but it is not currently known as a direct therapeutic drug target[1][4]. No drugs or specific clinical safety concerns associated with direct modulation of CCDC33 are reported.
