Overview

Coiled-coil domain-containing protein 34 (CCDC34) is a member of the large family of coiled-coil domain-containing proteins, notable for a structural motif that supports protein-protein interactions. CCDC34 is expressed highly in the testis and plays a critical role in the structural integrity of sperm flagella by maintaining the axonemal ultrastructure and the assembly or stability of outer dynein arms. Missense or frameshift mutations in CCDC34 cause severe sperm motility defects and are a genetic cause of male infertility (oligoasthenoteratozoospermia)[2]. CCDC34 is also overexpressed in various cancer cells, including hepatocellular carcinoma, where it has been implicated in regulating cell proliferation and migration. However, CCDC34 is not an enzyme, receptor, or traditional therapeutic drug target, and its detailed molecular mechanisms and broader physiological roles remain under active investigation[2].
