Coiled-coil domain-containing protein 38 (CCDC38) is a cytoskeletal protein characterized by the presence of a coiled-coil domain, which is a common structural motif that facilitates protein-protein interactions[1][3]. CCDC38 is coded by the CCDC38 gene (HGNC: 26843; NCBI Gene: 120935; UniProt: Q502W7) and is critical for male fertility as it is essential for sperm flagellum biogenesis and acrosome formation during spermiogenesis[5]. It is also required for the attachment of developing acrosomes to the nucleus and may help transport fibrous sheath components within sperm cells[5]. While its full cellular functions are not entirely defined, CCDC38 is thought to play a role in cilium and cytoskeleton-related processes and has been found expressed in bronchial epithelial cells, suggesting an extended involvement in lung function. A rare variant in CCDC38 has been associated with resistance to smoking-induced decline in lung function, potentially via its role in ciliary function[2]. CCDC38 itself has not been established as a direct therapeutic target, there are no known interacting drugs, and it is not recognized as a clinical biomarker or as presenting particular drug development safety concerns.
