Coiled-coil domain-containing protein 43 (CCDC43) is a member of the coiled-coil domain-containing protein family, characterized by structural motifs of two or more alpha-helices coiled together[1]. CCDC43 is a 224-amino-acid protein encoded on chromosome 17q21.31 and expressed in diverse tissues[1]. It is consistently upregulated in a variety of cancers, including lung, gastrointestinal, urogenital, and reproductive tract tumors, with its expression correlating with cell proliferation, invasion, metastasis, and decreased anti-tumor immunity[1][2][4]. Functional studies suggest roles in RNA or nucleic acid binding and multivalent protein interactions, with potential regulatory influence over cell death mechanisms and key signaling pathways (e.g., hippo, mTOR)[1][2]. CCDC43 may act upstream of mTORC1 and impact epithelial-mesenchymal transition, immune microenvironment modulation, and DNA repair processes in tumors. High CCDC43 expression portends poor prognosis and could help stratify patients for immunotherapeutic benefit[2][4]. Currently, there are no approved drugs specifically targeting CCDC43, but it is under investigation as a novel therapeutic and diagnostic target, especially in oncology.
