Overview

Coiled-coil domain-containing protein 50 (CCDC50) is a multifunctional signaling and adaptor protein encoded on chromosome 3, with high expression in neuronal cells and immune tissues[1][3][4]. It plays key roles in EGFR-mediated cell signaling, negatively regulates NF-κB signaling, and is directly involved in cell fate processes such as apoptosis, inflammation, and proliferation[1][5][6][7]. CCDC50 is essential in the clearance of damaged lysosomes as a lysophagy receptor, thereby supporting tumor cell survival and invasion by protecting against oxidative and lysosomal stress[2]. In the nervous system, CCDC50 is crucial for neuronal differentiation, axonal outgrowth, and may play a role in hearing via organization of the microtubule cytoskeleton in cochlear hair cells[1][4][5]. Mutations in this gene are causative for DFNA44, a form of hereditary hearing loss, while its overexpression promotes tumor growth. CCDC50 has no known direct drug interactions but is a potential context-dependent therapeutic target in oncology, autoimmunity, and neurology.
