Coiled-coil domain-containing protein 54 (CCDC54) is a structural protein that plays a key role in the architecture of the spermatozoa flagellar fibrous sheath and is essential for sperm motility and function. It is widely expressed in testicular tissue and sperm, with additional expression reported in somatic and cancer cells, suggesting possible regulatory roles in cell adhesion, migration, and tumorigenesis. Its sequence features a coiled-coil domain and, in certain homologs, a calmodulin-binding motif and carbohydrate-binding region, underlying its multifaceted cellular interactions. While not an established therapeutic target, CCDC54 and its close homolog SPA17 (often referred to interchangeably as SP17 or NYD-SP17) are under investigation for their roles in reproductive biology and cancer, with potential as biomarkers for diseases involving aberrant cell adhesion or motility. However, disease mechanisms and therapeutic relevance remain areas of ongoing research. Important caveat: There is confusion in the literature between the genes/proteins CCDC54 and SPA17, both of which are involved in sperm biology and sometimes both referred to as "SP17" or "NYD-SP17". Careful gene/protein mapping is recommended before making any therapeutic claims. If you are interested in the "SP17" as referenced in most cancer or testis literature, you may wish to consult sources on the SPA17 gene. If you need structured data for both CCDC54 and SPA17 separately, confirmation of context will be needed, as only SPA17/SP17 is directly linked to the sperm zona pellucida adhesion and most cancer/biomarker studies.
