Overview

Coiled-coil domain-containing protein 57 (CCDC57) is a multifunctional human protein encoded by the CCDC57 gene located on chromosome 17. It acts as a pleiotropic regulator of centriole duplication, mitosis, and ciliogenesis. CCDC57 localizes to the proximal end of centrioles in the centrosome and interacts with key proteins such as CEP63 and CEP152, which are essential for accurate centriole duplication and mitotic progression. It also binds microtubules and centriolar satellites, coordinating processes that ensure proper cell division and cilium assembly. Loss of CCDC57 disrupts these critical events, potentially contributing to genomic instability, cancer, developmental disorders like primary microcephaly, and defects in cell cycle regulation[1][2][4][6]. CCDC57 is not currently known as a therapeutic target, nor are there any drugs or well-validated biomarkers associated with its modulation.
