Overview

Coiled-coil domain-containing protein 66 (CCDC66) is a highly conserved microtubule-associated protein that regulates cilium assembly, maintenance, and disassembly and is required for proper ciliary signaling and epithelial organization[1][4]. CCDC66 localizes to the ciliary axoneme, centrosome, centriolar satellites, and spindle apparatus during mitosis, where it regulates microtubule organization and nucleation, as well as actin cytoskeleton interactions[1][2][3][4][5]. Mutations or loss of CCDC66 are linked to defects in ciliary function, ciliopathies such as Joubert syndrome, and retinal degeneration, highlighting its essential role in cytoskeletal processes and cell division[1][4][5]. There are no known small-molecule drugs or therapeutic modulators currently reported to interact specifically with CCDC66 in a clinical setting[4].
