Coiled-coil domain-containing protein 68 (CCDC68) is a centrosomal protein essential for centriole subdistal appendage assembly and microtubule anchoring in interphase cells[7][6][3]. It is part of the centriolar subdistal appendage, cooperating with other components such as CCDC120, ODF2, NIN, and CEP170 for hierarchical assembly[7]. Biologically, CCDC68 is involved in protein localization and cell cycle regulation, and it has a context-dependent role in cancer: it functions as a tumor suppressor in human colorectal cancer and pancreatic ductal adenocarcinoma, where its downregulation correlates with increased cell proliferation and worse prognosis; conversely, in non-small cell lung cancer, CCDC68 is upregulated and appears to promote cell proliferation[1][2][4]. Mechanistically, tumor-suppressive effects in colorectal cancer involve cell cycle arrest in G0/G1 via a CDK4 downregulation and increased CDK4 protein degradation through the RXRα/ITCH pathway[1]. The gene is also linked to other syndromes, such as recurrent pregnancy loss and lymphoma, but no direct drug interactions, mechanisms of drug action, or safety concerns are established[3].
